The easiest/cheapest way is to take a bus from the Istanbul Otogar to Bodrum. It will take 10-12 hours by bus, but Turkish buses are quite nice and depending on your budget, they are much cheaper than traveling by plane. Good bus companies: VARAN or KAMIL KOC. You could also fly from Istanbul to Bodrum, but it will cost you more. Check Turkish Airlines, Onur Airlines, Atlas Jet for fares. They have websites but are sometimes difficult to navigate.
It’s difficult to plan these kinds of things online, actually. If you are flying into Istanbul I recommend that you contact a travel agent who can help you organize bus or plane tickets within Turkey.

Car would take 8-10 hrs. I was in Bodrum by a car, not trough Istanbul but close enough to tell you. Hope this helps
